目的:研究基质金属蛋白酶2(MMP-2)在胰腺癌组织中表达及其与增殖细胞核抗原(PCNA)的相关性。方法:应用组织芯片制作仪构建胰腺癌组织芯片,在连续的芯片切片上使用免疫组化法检测MMP-2和PCNA蛋白表达情况,分析MMP-2与胰腺癌临床病理因素之间的关系以及与PCNA表达的相关性。结果:MMP-2在胰腺癌组织中表达的阳性率为66.7%(28/42),明显高于正常胰腺组织的10.3%(4/39),P<0.05。MMP-2表达与胰腺癌的分化程度、淋巴结转移以及分期相关(P<0.05),并且与PCNA的表达存在显著的相关性(P<0.01)。结论:MMP-2蛋白在增殖能力强的胰腺癌细胞中表达明显增加,与胰腺癌的转移有着密切关系。
Objective: To investigate the expression of matrix metalloproteinase 2 (MMP-2) in pancreatic cancer and its relationship with proliferating cell nuclear antigen (PCNA). Methods: The tissue microarray was used to construct pancreatic cancer tissue microarray. The expression of MMP-2 and PCNA protein was detected by immunohistochemistry on the sections of serial chips. The relationship between MMP-2 and clinicopathological factors of pancreatic cancer was analyzed. Correlation of PCNA expression. Results: The positive rate of MMP-2 expression in pancreatic cancer tissues was 66.7% (28/42), significantly higher than that in normal pancreatic tissues (10.3%, P <0.05). The expression of MMP-2 was correlated with the differentiation, lymph node metastasis and staging of pancreatic cancer (P <0.05), and there was a significant correlation between the expression of MMP-2 and PCNA (P <0.01). Conclusion: The expression of MMP-2 protein is significantly increased in pancreatic cancer cells with strong proliferative ability, which is closely related to the metastasis of pancreatic cancer.
