以地枫皮为试材,采用正交实验研究了无机盐、生长调节素、渗透压等对地枫皮离体保存的影响,建立地枫皮种质资源离体保存技术体系;并通过生长恢复的考察对保存材料进行初步评价。结果表明:无机盐、生长调节剂、渗透压等因素对地枫皮种质资源的离体保存均有显著影响,最终确定地枫皮常温离体保存的最佳保存方法为1/2MS+蔗糖50g/L+琼脂4.0g/L+甘露醇5g/L+矮壮素(CCC)1.0mg/L,培养条件为光照时间12~14h/d,培养温度25±2℃,光照强度2 000lx,在此条件下保存300d,存活率在50%以上,保存材料生长恢复情况良好。
