目的:本实验旨在研究大鼠运动至力竭过程中,大豆总皂苷(TS)对心肌抗氧化能力的影响。方法:用跑台一次运动力竭模型,将受试大鼠分为4组:对照组、TS组、力竭组、TS力竭组。按每千克体重20毫克、每天一次的剂量持续两周喂服TS组和TS力竭组大鼠。力竭组喂服安慰剂,对照组不给予任何处理。将跑步机速度预设为30 m/min,让力竭组和TS力竭组大鼠进行跑台运动直至力竭,立即处死并制备10%的心肌组织匀浆。结果:与力竭组相比,TS力竭组运动时间显著延长20.62%(p<0.05)。与对照组相比,TS组CAT、GSH-Px和GR酶活性显著增强(p<0.01),TS力竭组GSH-Px与GR酶活性显著增强(p<0.01)而MDA水平显著降低(p<0.05)。与力竭组相比,TS力竭组GSH-Px活性显著增强(p<0.01),TS组CAT、GSH-Px和GR酶活性均显著提高(p<0.01)。与TS组相比,TS力竭组CAT和GR活性显著增强(p<0.01)。结论:TS可不同程度地改善运动大鼠的心肌抗氧化活性,降低大鼠心肌MDA水平、血清AST和LDH水平,延长运动时间并延缓运动疲劳的发生。
OBJECTIVE: The purpose of this study was to investigate the effects of total soy saponin (TS) on myocardial antioxidant capacity during exercise to exhaustion in rats. Methods: The rats were divided into 4 groups: control group, TS group, exhaustion group and TS exhaustion group by using treadmill exercise model. At a dose of 20 mg per kg of body weight once daily, rats in TS group and TS-exhausted group were fed for two weeks. Exhaustion group fed placebo, the control group did not give any treatment. The treadmill speed was pre-set to 30 m / min, so that exhaustive and TS exhausted rats were treadmill running until exhaustion, immediately sacrificed and prepared 10% of myocardial tissue homogenate. Results: Compared with the exhaustive group, exercise time in the TS exhausted group was significantly prolonged by 20.62% (p <0.05). Compared with the control group, the activities of CAT, GSH-Px and GR in TS group were significantly increased (p <0.01), the activity of GSH-Px and GR enzyme in TS group was significantly increased (p <0.01) <0.05). Compared with the exhaustive group, the activity of GSH-Px in TS-depleted group was significantly increased (p <0.01), while the activities of CAT, GSH-Px and GR in TS group were significantly increased (p <0.01). Compared with TS group, the activity of CAT and GR in TS depletion group was significantly increased (p <0.01). CONCLUSION: TS can improve myocardial antioxidant activity, decrease myocardial MDA, serum AST and LDH, prolong exercise time and delay exercise-induced fatigue in exercising rats.
