目的:采用HPLC法研究比较黄柏丝及其不同炮制品的指纹图谱,以探讨黄柏的炮制机理。方法:以95%乙醇(含1%盐酸)为提取溶剂,采用超声方法提取制备供试样品。以Kromasil-C18色谱柱为固定相,甲醇:磷酸缓冲液(0.2%磷酸用三乙胺调节pH值至3.55)为流动相进行梯度洗脱,进行HPLC指纹图谱检测分析。结果:盐黄柏与黄柏丝相比,主要色谱峰峰面积有所降低。黄柏炭与黄柏丝相比,指纹图谱发生了显著改变,多数主要色谱峰已基本检测不到。结论:本研究建立了能较好反映黄柏中化学成分的HPLC指纹图谱方法,可用于黄柏及其炮制品的质量控制。
OBJECTIVE: To study the fingerprints of the yellow cedar and its processed products by HPLC to investigate the processing mechanism of the cork. Methods: 95% ethanol (containing 1% hydrochloric acid) as extraction solvent, ultrasonic extraction method for the preparation of test samples. The mobile phase consisted of Kromasil-C18 as the mobile phase and methanol: phosphate buffer (0.2% phosphoric acid and triethylamine adjusted to pH 3.55) as the mobile phase. Results: Compared with yellow cedar, the main chromatographic peak area decreased. Compared with yellow cork, Cork’s carbon fingerprints have changed significantly, most of the major peaks have not been detected. Conclusion: This study established a HPLC fingerprinting method that can better reflect the chemical constituents of Phellodendron amurense, which can be used for the quality control of Phellodendron and its processed products.
