目的:观察中华眼镜蛇毒F组分对血小板α-颗粒膜蛋白表达的影响,确定F组分是否具有抑制血小板活化的功能。方法:实验分成对照组、凝血酶组和各给药组(凝血酶+F组分),应用酶联免疫吸附法(ELISA)测定健康成年人血浆中的α-颗粒膜蛋白(GMP-140)含量的变化。结果:凝血酶能引起血浆中的GMP-140含量明显升高,中华眼镜蛇毒F组分 100、30 μg/mL明显降低凝血酶引起的血浆GMP-140含量升高,并呈现一定程度的剂量依赖关系,两组比较P<0.05。结论:中华眼镜蛇毒F组分能够抑制激动剂引起的血小板活化。
OBJECTIVE: To observe the effect of F component of Naja naja atra Venom on platelet α-granule membrane protein expression and determine whether F component has the function of inhibiting platelet activation. Methods: The experiment was divided into control group, thrombin group and each administration group (thrombin + F component). The content of α-granule membrane protein (GMP-140) in plasma of healthy adults was determined by enzyme-linked immunosorbent assay (ELISA). The change. RESULTS: Thrombin could cause a significant increase in plasma GMP-140 content. The 100, 30 μg/mL F fraction of Chinese cobra venom significantly reduced thrombin-induced plasma GMP-140 levels and showed a dose-dependent effect. The relationship between the two groups was P<0.05. Conclusion: The F component of Naja naja atra can inhibit agonist-induced platelet activation.
