在PHA(1μg/ml)刺激下,IL-1或TNF均能诱发静止T细胞发生增殖反应,其最适浓度为IL-1(50U/ml)和TNF(200U/ml)。利用其最适浓度,联合作用于PHA刺激T细胞,其增殖反应增强了25%,但仍低于PHA联合10%的Mφm所诱发的增殖反应。单克隆抗体anti-Tac(p55)完全阻断了由PHA联合IL-1所诱发的增殖反应,但对PHA联合TNF所诱发的增殖反应只能部份地抑制。这些结果表明,TNF除了同IL-1一样具有通过IL-2途径来诱导T细胞活化外,还有着其他的途径,提示T细胞活化增殖的第二信号可由多种单核因子提供,而最大的增殖反应的发生,则是多种单核因子和其他因素协同作用的结果。
IL-1 or TNF could induce the proliferation reaction of resting T cells stimulated by PHA (1μg / ml) at the optimal concentration of IL-1 (50U / ml) and TNF (200U / ml). Using its optimal concentration, the proliferation of T cells stimulated by PHA was enhanced by 25%, but still lower than that of PHA combined with 10% Mφm. Monoclonal antibody anti-Tac (p55) completely blocked the proliferative response induced by PHA combined with IL-1, but only partially inhibited the proliferative response induced by PHA combined with TNF. These results indicate that TNF has other pathways besides inducing T cell activation through the IL-2 pathway as well as IL-1, suggesting that the second signal of T-cell activation and proliferation may be provided by a variety of mononuclear factors and the largest Proliferation reaction occurs, it is a variety of single-factor and other factors synergistic results.
