目的设计并合成3个新型近红外吲哚七甲川菁染料(Ⅲ1、Ⅲ2、Ⅲ3),并探讨其荧光成像的活性。方法将6-溴己酸与取代苯酚成酯后,再与2,3,3-三甲基-3H吲哚反应制备成季铵盐,进而与环己烯双醛缩合制得目标化合物;通过测定目标物的最大吸收波长、发射波长以及评价体外近红外成像的特点,选择最优化合物进行动物活体成像测试。结果与结论合成并确证了目标物的化学结构,其最大吸收和发射波长均在近红外区;小鼠活体成像研究显示:吲哚七甲川菁染料Ⅲ1可用于体内组织器官的近红外显像。
Objective To design and synthesize three novel near infrared indole heptamethyocyanine dyes (Ⅲ 1, Ⅲ 2, Ⅲ 3) and investigate their fluorescence imaging activities. Methods The 6-bromohexanoic acid was esterified with substituted phenol and then reacted with 2,3,3-trimethyl-3H-indole to prepare a quaternary ammonium salt, which was then condensed with cyclohexene dialdehyde to obtain the target compound. Determination of the maximum absorption wavelength of the target, the emission wavelength and the evaluation of the characteristics of near-infrared imaging in vitro, select the optimal compound for animal biopsy test. RESULTS AND CONCLUSIONS The chemical structure of the target was synthesized and confirmed. The maximum absorption and emission wavelengths of the target were in the near infrared region. In vivo imaging of mice showed that indole-1,7-methylene blue dye Ⅲ could be used for near-infrared imaging of tissues and organs in vivo.
