目的观察中药健脑方对脑缺血后大鼠海马齿状回神经发生以及空间学习记忆功能的影响。方法SD大鼠随机分为假手术组、模型组和治疗组。采取永久结扎双侧颈总动脉制作全脑缺血模型。术后治疗组给予健脑方灌胃干预,模型组给予等量生理盐水。使用BrdU标记技术观察缺血后海马齿状回的神经发生,采用Morris水迷宫试验评价大鼠空间学习能力。结果模型组和治疗组海马齿状回BrdU阳性细胞数分别在缺血后第7天达到最高峰。而缺血后7、14、20、28 d治疗组BrdU阳性细胞数明显多于模型组(P<0.05或P<0.01)。水迷宫试验显示术后第14天治疗组大鼠寻台平均时间明显短于模型组(P<0.05)。结论健脑方能持续明显促进脑缺血后大鼠海马齿状回神经发生,并改善大鼠的空间学习能力。
Objective To observe the effect of Chinese medicine Jiannao recipe on neurogenesis and spatial learning and memory function of hippocampal dentate gyrus in rats after cerebral ischemia. Methods SD rats were randomly divided into sham operation group, model group and treatment group. A permanent cerebral ischemia model was established by permanent ligation of both common carotid arteries. The treatment group was given intragastric administration of Jiannao Fang and the model group was given the same amount of normal saline. The neurogenesis of hippocampal dentate gyrus after ischemia was observed using BrdU labeling technique. Morris water maze test was used to evaluate the spatial learning ability of rats. Results The number of BrdU-positive cells in the dentate gyrus of the hippocampus in the model group and the treatment group peaked at 7 days after ischemia respectively. The number of BrdU-positive cells in the treatment group was significantly higher than that in the model group on the 7th, 14th, 20th, and 28th days after ischemia (P<0.05 or P<0.01). The water maze test showed that the mean time for seeking to stand in the treatment group on the 14th day after operation was significantly shorter than that in the model group (P<0.05). Conclusion Jiannao Prescription can significantly promote the neurogenesis of dentate gyrus in hippocampus of rats after cerebral ischemia, and improve the spatial learning ability of rats.
