目的:观察活血化瘀利水消肿方对大鼠桡骨骨折愈合的影响。方法:取成年SD大鼠90只,大鼠双侧桡骨骨折造模,造模后大鼠随机分为9组,活血化瘀利水消肿方组(总称A组)、活血化瘀方组(总称B组)、模型组(总称C组)各Ⅰ、Ⅱ、Ⅲ组。A、B、C组分别予活血化瘀利水消肿方煎剂、活血化瘀方煎剂、蒸馏水灌胃3周。造模后28、35、42d处死各Ⅰ、Ⅱ、Ⅲ组大鼠,取出大鼠桡骨做生物力学测试、BMP-2免疫组化观察。造模后当天、28、35、42d各组大鼠拍双侧桡骨X线片。结果:经过灌胃治疗,活血化瘀利水消肿方组大鼠桡骨的生物力学测试、X线评价优于模型组(P<0.05,P<0.01),BMP-2染色程度在造模后28、35d均高于模型组(P<0.05,P<0.01)。结论:活血化瘀利水消肿方能提高骨组织中BMP-2的含量,促进成骨细胞的生成,加快骨痂形成,增强骨组织的强度、韧性,缩短骨折愈合的时间。
Objective: To observe the effect of Promoting Blood Circulation and Removing Blood Stasis and Eliminating Dampness and Dissipating Recipe on the healing of radius fracture in rats. Methods: 90 adult Sprague-Dawley rats were used to make bilateral radius fracture models. After modeling, the rats were randomly divided into 9 groups: Huoxue Huayu Lishui Decoction (group A), Huoxue Huayu Recipe Collectively referred to as group B), model group (collectively C group) of each Ⅰ, Ⅱ, Ⅲ group. Groups A, B and C were given decoction for promoting blood circulation to remove stasis and diuresis, decoction for promoting blood circulation and removing blood stasis, and distilled water for 3 weeks respectively. At 28, 35 and 42 days after operation, all rats in group Ⅰ, Ⅱ and Ⅲ were sacrificed and the radius of the rats was removed for biomechanical test and BMP-2 immunohistochemistry. The day after modeling, 28,35,42 d of each group were taken on both sides of radial radiographs. Results: After gavage treatment, the biomechanical test and X-ray evaluation of the radial bone in the Huoxue Huayu liaozhengfang group were better than those in the model group (P <0.05, P <0.01). The staining level of BMP- , 35d higher than the model group (P <0.05, P <0.01). CONCLUSION: The activity of promoting blood circulation and reducing blood stasis can increase the content of BMP-2 in bone tissue, promote the formation of osteoblasts, accelerate the formation of callus, increase the strength and toughness of bone tissue and shorten the healing time of fractures.
