目的采用基因芯片高通量研究浙江金华地区人群乙醛脱氢酶2(ALDH2)基因多态性及生活习惯与胃癌易感性,旨在为本地区胃癌预防提供参考。方法选取胃癌患者161例,设为胃癌组;另采用配对病例对照研究设计方案,选取健康志愿者161例,设为对照组。采用调查问卷采集受试者信息,采集血样检测ALDH2基因多态性,分析ALDH2基因多态性及生活习惯与胃癌易感性的关系。结果胃癌组ALDH2基因型中,野生型纯合子(G/G)、杂合子(G/A)和变异型纯合子(A/A)分别占64.60%、31.06%、4.34%,对照组分别为61.49%、37.27%、1.24%,胃癌组与对照组基因型比较,差异均无统计学意义(P>0.05)。ALDH2G/A和A/A基因型并未增加罹患胃癌的危险性。携带ALDH2G/A或A/A者与携带ALDH2G/G者相比,饮酒、吸烟、常食用腌制食品、三餐不定时、暴饮暴食等不良生活习惯均导致罹患胃癌的风险增加。结论浙江金华地区人群ALDH2G/A和A/A基因型并未增加罹患胃癌的危险性,饮酒、吸烟、常食用腌制食品、三餐不定时、暴饮暴食等不良生活习惯均是胃癌的危险因素,由此推测胃癌的发生发展可能是遗传与环境因素共同作用下的多步骤、多阶段的连锁效应过程。
Objective To study the genetic polymorphism of aldehyde dehydrogenase 2 (ALDH2) gene and lifestyle and susceptibility to gastric cancer using high-throughput microarray in Jinhua, Zhejiang Province, and to provide references for the prevention of gastric cancer in this area. Methods A total of 161 patients with gastric cancer were enrolled in this study. The patients were divided into gastric cancer group and control group. 161 healthy volunteers were selected as the control group. Subjects were collected by questionnaire, blood samples were collected to detect ALDH2 gene polymorphism, ALDH2 gene polymorphism and lifestyle and gastric cancer susceptibility were analyzed. Results The percentage of wild type homozygotes (G / G), heterozygotes (G / A) and variant homozygotes (A / A) accounted for 64.60%, 31.06% and 4.34% respectively in ALDH2 genotype of gastric cancer group, 61.49%, 37.27%, 1.24% respectively. There was no significant difference in the genotype between gastric cancer group and control group (P> 0.05). ALDH2G / A and A / A genotypes did not increase the risk of gastric cancer. Patients with ALDH2G / A or A / A, compared with those with ALDH2G / G, had an increased risk of developing gastric cancer due to alcohol, smoking, regular consumption of preserved foods, occasional meals, overeating and other unhealthy habits. Conclusions ALDH2G / A and A / A genotypes in Jinhua, Zhejiang Province did not increase the risk of gastric cancer. Alcohol consumption, smoking, regular consumption of preserved foods, irregular meals, overeating and so on were the risk of gastric cancer It is speculated that the occurrence and development of gastric cancer may be multi-step and multi-stage chain reaction under the combined effect of genetic and environmental factors.
