目的 :探讨喉鳞状细胞癌 (LSCC)组织中血管内皮生长因子 (VEGF)mRNA及蛋白的表达和微血管密度 (MVD)与颈淋巴结转移的关系。方法 :用半定量逆转录 聚合酶链式反应 (semi RT PCR)技术和免疫组织化学方法检测 6 0例LSCC组织中VEGFmRNA及蛋白的定量表达及MVD。结果 :VEGF的表达无论在基因水平还是蛋白水平伴有颈淋巴结转移组都显著高于不伴有颈淋巴结转移组 (P <0 .0 1)。CD31免疫组织化学染色结果显示伴颈淋巴结转移组的MVD计数显著高于不伴颈淋巴结转移组 (P <0 .0 1)。LSCC组织中MVD计数升高与VEGF高表达呈正相关 (r =0 .94 8 4 ,P <0 .0 5 ) ,且二者与颈淋巴结转移相关。结论 :VEGF表达和瘤内MVD有良好的相关性 ,提示VEGF高表达对LSCC微血管生成起重要作用 ;VEGF高表达和瘤内高MVD与LSCC的颈淋巴结转移密切相关 ,二者有可能成为预测LSCC转移和预后的生物学指标 ;抗VEGF及其受体的抗血管生成治疗可能成为LSCC治疗的靶点
Objective: To investigate the relationship between the expression of vascular endothelial growth factor (VEGF) mRNA and microvessel density (MVD) and cervical lymph node metastasis in laryngeal squamous cell carcinoma (LSCC). Methods: The semi-quantitative reverse transcriptase-polymerase chain reaction (semi RT PCR) and immunohistochemistry were used to detect the expression of VEGF mRNA and protein and MVD in 60 LSCC tissues. Results: The expression of VEGF was significantly higher in patients with cervical lymph node metastasis than those without cervical lymph node metastasis (P <0.01). The results of CD31 immunohistochemical staining showed that the MVD counts in patients with cervical lymph node metastasis were significantly higher than those without cervical lymph node metastasis (P <0.01). The increase of MVD in LSCC was positively correlated with the high expression of VEGF (r = 0.9484, P <0.05), and both were correlated with lymph node metastasis. CONCLUSIONS: The correlation between the expression of VEGF and MVD in the tumor is good, which indicates that the high expression of VEGF plays an important role in the angiogenesis of LSCC. The high expression of VEGF and the high MVD in the tumor are closely related to the cervical lymph node metastasis of LSCC. Metastasis and prognosis of the biological indicators; anti-VEGF and its receptor anti-angiogenic therapy may be the target of LSCC treatment
