目的探讨研究凝血酶敏感蛋白-1(TSP-1)对胆囊癌细胞株GBC-SD生长增殖的作用及其作用途径。方法根据处理因素将胆囊癌细胞分为:空白对照组,TSP-1组,CD36单抗阻断组、CD47单抗阻断组。用四唑盐比色法(MTT)和流式细胞术检测TSP-1及受体CD36、CD47对胆囊癌细胞生长、细胞周期的影响。结果TSP-1组、CD36阻断组、CD47阻断组细胞生长明显低于对照组(抑制率分别为:23.63%,9.57%,18.13%)。CD36阻断组的抑制率小于CD47阻断组。TSP-1组G0/G1期细胞数增加,S期和G2/M期细胞数减少,与对照组和CD36阻断组相比有显著性差异。增殖指数分别为:对照组(39.55±2.12)、TSP-1组(26.21±0.61)、CD36阻断组(36.43±1.28)、CD47阻断组(30.73±0.38);各组之间两两比较均有差异。结论TSP-1可对抑制胆囊癌细胞的生长,通过与受体CD36的结合阻滞胆囊癌细胞停止于G0/G1期可能是主要的作用途径。
Objective To investigate the effect of thrombin-sensitive protein-1 (TSP-1) on the growth and proliferation of gallbladder carcinoma cell line GBC-SD and its action pathway. Methods Gallbladder cancer cells were divided into blank control group, TSP-1 group, CD36 monoclonal antibody blocking group and CD47 monoclonal antibody blocking group according to the treatment factors. The effects of TSP-1, CD36 and CD47 on the growth and cell cycle of gallbladder cancer cells were detected by MTT assay and flow cytometry. Results The cell growth of TSP-1 group, CD36 blocking group and CD47 blocking group was significantly lower than that of the control group (inhibition rates were 23.63%, 9.57%, and 18.13%, respectively). The inhibition rate of the CD36 blocking group was lower than that of the CD47 blocking group. The number of cells in the G0/G1 phase of the TSP-1 group increased, the number of cells in the S phase and the G2/M phase decreased, and there was a significant difference compared with the control group and the CD36 blocking group. Proliferation indexes were: control group (39.55±2.12), TSP-1 group (26.21±0.61), CD36 blocking group (36.43±1.28), and CD47 blocking group (30.73±0.38); There are differences. Conclusion TSP-1 can inhibit the growth of gallbladder cancer cells, and the arrest of gallbladder cancer cells in G0/G1 phase through the binding with receptor CD36 may be the main action pathway.
