目的:以痔疮患者肠道内IL-17、IL-6、TNF-α细胞因子为研究对象,检测其在痔疮患者中的表达,探讨细胞因子在痔疮发病机制中的作用。方法:采用免疫组化方法测定痔疮患者病变部位直肠黏膜和血管内皮细胞内IL-17、IL-6、TNF-α表达。酶联免疫吸附试验(ELISA)方法检测痔疮患者(试验组)与对照组血清IL-17蛋白表达。结果:试验组与对照组血清IL-17表达无差异(P>0.05);试验组与对照组直肠血管内皮细胞IL-17、IL-6、TNF-α表达有显著差异(P<0.05);试验组直肠黏膜IL-17、IL-6、TNF-α表达与对照组相比也都具有显著性差异(P<0.05)。结论:据此推测局部阻断IL-17、IL-6、TNF-α等细胞因子的分泌可能成为治疗痔疮的一种方法。
Objective: To study the expression of IL-17, IL-6 and TNF-α in the intestine of patients with hemorrhoids and to investigate the role of cytokines in the pathogenesis of hemorrhoids. Methods: The expressions of IL-17, IL-6 and TNF-α in rectal mucosa and vascular endothelial cells of hemorrhoids patients were determined by immunohistochemistry. Serum IL-17 protein expression in hemorrhoid patients (experimental group) and control group was detected by enzyme-linked immunosorbent assay (ELISA). Results: There was no significant difference in serum IL-17 expression between experimental group and control group (P> 0.05). The expressions of IL-17, IL-6 and TNF-α in rectal vascular endothelial cells of experimental group and control group were significantly different (P <0.05) The expression of IL-17, IL-6 and TNF-α in rectal mucosa of experimental group also had significant difference compared with control group (P <0.05). Conclusions: It is speculated that local block of IL-17, IL-6, TNF-α and other cytokines secretion may become a method of treatment of hemorrhoids.
