目的研究长春市双阳区鼠类中汉坦病毒感染情况及其基因分型。方法收集2011年双阳地区捕获鼠类,取鼠肺组织,用间接免疫荧光检测,阳性标本进行汉坦病毒M片段扩增并测序,与其他毒株进行相似性比对及系统树分析。结果收集100份鼠肺,经IFA检测到1份阳性,阳性率为1%,对其进行逆转录巢式PCR扩增,结果为汉城(SEO)型HV,与代表株进行相似性比较发现,SY-4部分M基因片段序列与其他SEOV核苷酸相似性为95.4%~99.7%,氨基酸序列相似性为99%~100%。系统树显示,该序列属S3亚型。结论本次获得HV是SEO型,亚型为S3亚型。
Objective To study the prevalence and genotyping of Hantaviruses in mice in Shuangyang District of Changchun City. Methods The murine rats were collected from rats in Shuangyang in 2011. The positive samples were detected by indirect immunofluorescence. The Hantaan virus M fragments were amplified and sequenced. The similarity and phylogenetic tree were also compared with other strains. Results A total of 100 rat lungs were collected and 1 positive by IFA. The positive rate was 1%. Reverse transcriptional nested polymerase chain reaction (PCR) The sequence of SY-4 part M gene shares 95.4% -99.7% similarity with other SEOV nucleotides and the amino acid sequence similarity is 99% -100%. The phylogenetic tree shows that this sequence is of the S3 subtype. Conclusions This study found that HV is SEO type and subtype is S3 subtypes.
