黄瓜花叶病毒(Cucumber mosaic virus, CMV)在番茄上造成的危害十分严重,可严重降低番茄的产量和质量。本试验利用人工microRNA(artificial microRNA, amiRNA)技术使转化的番茄具有对CMV的抗性。amiRNA技术在我国番茄抗CMV的育种研究中尚未见报道。将人工改造的植物microRNA(miRNA)转入番茄体内,获得的转化番茄,对番茄抗病毒育种研究和miRNAs的研究与应用具有重要意义。本试验首先分析番茄中的miRNAs本底表达量,选择表达量较高的miRNA159作为人工构建miRNA的骨架。使用siRNA Target Finder设计靶标基因为CMV2a基因的的siRNA,并根据BLAST结果选择一段与番茄同源性最低的片段,用其替换miRNA159的成熟序列,并将amiRNA159-2a连接于载体pBI121 ,获得重组载体pBI121-amiRNA159-2a。通过农杆菌EHA105的介导将重组载体pBI121-amiRNA159-2a,转入番茄体内。为了提高转化的效率,对转化体系的分化出芽激素、卡那霉素、脱菌素和生根激素等进行筛选,建立了优化的转amiRNA159-2a番茄体系。将得到的经过卡那霉素筛选的转amiRNA159-2a番茄再生苗进行分子鉴定,分别用PCR扩增和Southern杂交进行分子鉴定,并获得阳性植株。获得阳性转化子后,转amiRNA159-2a番茄的再生苗接种Fny-CMV,进行抗病性鉴定,结果表明转amiRNA159-2a再生苗比非转amiRNA159-2a植株明显抗CMV侵染。本研究成功构建了作用于CMV2a基因的重组载体pBI121-amiRNA159-2a;成功将amiRNA159-2a转入番茄体内,获得转amiRNA159-2a番茄植株;研究表明转amiRNA159-2a番茄具有明显的抗CMV特性。
