Trypsin is currently the most widely adopted protease for protein digestion in shotgun proteomic studies.Though effectively used both in the digestion of simple and complex samples, there are still some limitations in traditional in-solution digestion approach , such as autodigestion, long incubation time and non-reusability of the enzyme.Therefore, immobilized trypsin digestion is emerging as a fast, automated and highly efficient alternative.It can increase the efficiency of the protein digestion and shorten the digestion time from 16h to lmin.As the result, protein digestion using enzyme immobilized on the inner wall of capillary column is attracting more and more attention due to its advantages, such as easy preparation, on-line manipulation and high efficiency.
