  目的:为弄清辐射诱导IER5基因的转录调控机制.方法:采用生物信息学分析技术和缺失体构建、定点突变、EMSA及ChIP生物学实验方法,用宫颈癌hela细胞,确定IER5基因启动子区域、转录因子和结合位点.结果:实验发现IER5基因启动子最可能的较小范围为-408bp至-238bp.IER5基因具有两个转录因子,分别为GCF、NFI,其中GCF具有两个结合位点,分别位于在IER5基因启动子-388 bp ~-382 bp和-274 bp ~-270 bp处;NFI具有一个结合位点,该结合位点位于IER5基因启动子-362 bp ~-357 bp.结论:这两种转录因子对IER5基因的调控作用正相反,转录因子GCF对顺式作用元件起负性调节作用,并随照射剂量的增加而减弱;转录因子NFI对顺式作用元件起正性调节作用.
