玉米大斑病(northern corn leaf blight)是一种由大斑刚毛球腔菌(Setosphaeria turcica)引起的严重威胁玉米生产的真菌性病害。本课题组前期研究发现,cAMP信号途径对玉米大斑病菌的营养菌丝生长及致病性具有重要的调控作用,PKA作为该信号途径的下游调控因子,由两个催化亚基(PKA-C)和两个调节亚基(PKA-R)构成,针对该途径进一步从分子水平解析病原菌致病性的调控机制,将为寻找高效、持久的病害防控策略及发掘杀菌剂设计的新靶点奠定理论基础。本课题组前期利用RNAi技术获得沉默转化子,分析结果显示PKA调控病菌的附着胞发育及胞外水解酶活性影响其侵染能力,但其调控机制有待深入探索。因此,对玉米大斑病菌StpkaC1基因进行PEG介导的原生质体转化技术,经潮霉素抗性筛选、PCR、Southern bloting验证得到2株敲除转化子。对StpkaC1基因敲除突变菌株的形态及致病力进行分析,结果显示,与野生型相比,突变菌株生长速率加快,菌落颜色变浅,气生菌丝较多,且出现产孢缺陷,附着胞形成及穿透时间延迟,黑色素产量明显低于野生型,且致病力下降。以上结果表明,StpkaC1基因负调控菌株营养生长,对孢子产生、附着胞形成及穿透、黑色素合成均起正调控作用,并且调控菌株对寄主的致病力。
