Cyclospora spp.has been identified as one of the most important intestinal pathogens causing protracted diarrhea in animals and human beings. To investigate the prevalence and identify the species of Cyclosporain dairy cattle,a total of 2199 fecal samples from 15 different farms in Henan Province were collected and examined by using Sheater s sugar flotation technique. The DNA of all the samples was extracted and amplified the 18S rDNA partial sequence.A new restriction fragment length polymorphism (RFLP) enzyme BSpE Ⅰ was used to differentiate the Cyclospora species from cattlE-associated Eimeria spp.,the positive samples were confirmed by sequence. The results demonstrate that 42 positive samples were found, the ratio was 1.89%.Otherwise,the highest ratio infection in the age groups is 31:60d,is 4.50% (8/82),which is significant difference with other age groups.Phylogenetic analysis based on the 18S rDNA sequences revealed that the Cyclospora isolates from dairy cattle were posited into the Cyclospora sp.Guangzhou 1 (DQ082866.1) and Cyclospora sp.Oita (Ab73560-3.1).This is the first reported about the epidemiological of the big amount Cyclospora spp.infection in dairy cattle, which would not only provide useful information for further understanding of the molecular epidemiology of Cyclospora spp.,but also for the control ofCyclospora sp.infection in dairy cattle.
