Effects of hydrogels composite grafted with IKVAV on neural stem cells

  Successful stem cell-based therapy for neurological disorders is highly dependent on the survival of transplanted stem cells.We investigated the use of an injectable hydrogel grafted with IKVAV poly (lactide-co-ethylene oxide-co-fumarate) for en-hancing the survival of transplanted neural stem ceils (NSCs).The gelation properties of the hydrogel were first characterized and its biocompatibility was evaluated.In order to identify the biological safety of IKVAV composite hydrogel in vivo,we investigated the survival and proliferation of NSCs by immunofluorescence staining method.Different concentrations of grafted IKVAV poly (lactide-co-ethylene oxide-co-fumarate) hydrogels were seeded with NSCs derived from SD newborn rats,and then NSC viability and apoptosis assays were performed.Results showed that the effects of hydrogel on NSC surviv-al and proliferation were in a concentration-and time-dependent manner.NSC proliferation was highest after culture with 100 μg/mL IKVAV grafted hydrogels for 72 hours.After 24-hour culture,there was no difference in the number of apoptotic cells between 100 μg/mL IKVAV hydrogel-treated and IKVAV-free hydrogel groups.Taken together,hydrogel scaffolds can in-crease the survival of NSCs and 100 μg/mL IKVAV grafted hydrogels are likely to be the optimal hydrogel composition for the survival of NSCs.
