In order to investigate the effect of Brn-4 on the differentiation of the neural stem cells into neurons,we transected the fimbria of hippocampi of adult SD rats,then the extracts from fimbria transected and intact hippocampi were respectively obtained after the rats were breeding 14 days.Simultaneously the NSCs isolated from the hippocampus of embryonic rat were expanded and divided into 3 groups as following:(1) transection group:DMEM/F12 medium containing the extract of the fimbria-transected hippocampi was added; (2) normal group:DMEM/F12 medium containing the extract of the normal hippocampi was added; (3) control group:DMEM/F12 medium without any extract was added.The NSCs were cultivated and the neurons differentiated from the NSCs were detected by immunofluorescence doublelabeling of Brn-4 and MAP-2 on the 14th day after culturing.
