Au纳米颗粒的制备与表面修饰及其作为基因载体的体外生物学特性检测

  目的:研究Au纳米颗粒的制备、PEI对金纳米粒的修饰及表征,并对其作为基因载体的体外生物学特性进行检测.为临床应用提供理论和实验依据.材料与方法:(1)Au纳米颗粒的制备:采用单步还原法,以柠檬酸三钠作为还原剂和稳定剂制备Au纳米颗粒.取1%的HAuCl4水溶液lml加入97ml水中油浴加热沸腾,然后迅速加入2ml 1%的柠檬酸三钠水溶液保持沸腾15分钟即可得到Au纳米颗粒.(2)用PEI对Au纳米粒进行表面修饰:将10mlHAuCl4水溶液(浓度为0.01g/ml))与10mlPEI水溶液(浓度0.02g/ml)混合磁粒搅拌15min,然后加入1%的柠檬酸三钠2ml,继续搅拌一个小时.溶液变为紫红色后,透析去除游离的PEI,得到表面修饰过的PEI-Au.用透射电子显微镜(TEM),对Au和PEI-Au纳米颗粒进行表征.
