Background and purpose: Methamphetamine(MA)is a highly addictive stimulant that affects to neurotoxicity of the brain.Changes of brain metabolites and brain volume in MA abusers have been documented. However, the relationship between the changes of brain metabolites at a selected location and the changes of brain volume at the corresponding locations has never been reported. Therefore,we aim to explore the association between the change of Magnetic Resonance Spectroscopy(MRS)in 4 voxel locations and the change of brain volume at the corresponding locations or region. Methods: Ten MA users(age=24.5±4.7),and 14 healthy control(HC)(age=22.71±3.36)were recruited to this study.Single voxel MRS was acquired in 4 voxel locations including frontal white matter(FWM),frontal gray matter(FGM),posterior gray matter(PGM)and Basal Ganglia(BG)and using point resolved spectroscopy(PRESS)pulse sequence for scanning. For the brain volume,T1-weighted high resolution images were acquired using steady state 3D FFE pulse sequence, and were analyzed by FreeSurfer Version 5.3.Results: Significant reduction of N-acetylaspartate per Glycerophosphocholine(NAA/Cho)at the left FWM and left BG were found in MA compared to the HC groups,p=0.049 and p=0.039 respectively. The volume reduction of the left FWM was paralleled to the significant reduction of NAA/Cho in MA.In addition, the volume reduction of the left BG and FGM were significantly correlated with the reduction of NAA/Cho at the same location,r=0.425; p=0.022 and r=0.327; p=0.048 respectively. Conclusions: The results of MRS in this study agreed well with other previous studies.Additional information from this study were the association between brain volumes and the brain MRS.
