The inhibition of phosphoinositide 3-kinase (PI3K) acting upstream of Akt, has been extensively reported to be valuable in the treatment of cancerous cell growth.Several cell-surface receptors, especially those linked to tyrosine kinases activate the class Ia PI3Ks.Mutated growth factor receptors, ras and PTEN loss lead to the over-activation of the PI3K pathway.Beyond oacology, the effects of Pl3Kgamma inhibitors have applications in other indications, such as rheumatoid arthritis and other inflammatory diseases.A new generation of (AE)tema Zentaris low molecular weight pyridopyrazine compounds has been identified as highly potent and selective inhibitors of PI3K and dual inhibitors of PI3K and ERK.These new PI3K-inhibitors were profiled in biochemical and cellular assays and investigated in several efficacy mouse xenograft cancer models.Mode-of-action studies showed that this substance class acts as ATP competitive compounds.AEZS-126 was selected as candidate compound for further profiling and preclinical development.
