The goal of this study was to explore a novel source of hyperthermophilic β-galactosidase (lactase) which may be useful for the hydrolysis of lactose and production of galacto-oligosaccharides.A gene for β-galactosidase(PF0356) was cloned from the Pyrococcus furiosus DSM3638 genomic DNA.The gene was composed of 1452 bp encoding 483 amino acids with a predicted molecular mass of 57,000 Da.The gene was ligated to pET28(a) vector and expressed in E.coli.The enzyme was purified by heat treatment (80℃),along with Ni-NTA affinity chromatography.The purified β-galactosidase was composed as a dimer.
