基于调控机制构建雷帕霉素高产基因工程菌株

利用生物信息学同源性分析吸水链霉菌基因组中链霉菌属抗生素合成调控基因,发现存在正调控基因afsR及分化负调控基因nsdA高度同源序列.全基因合成吸水链霉菌类afsR、rapG、rapH等正调控基因、类nsdA负调控基因序列,分别连接到pSET152或pKC1139表达载体,CaCl2法转入大肠杆菌ET12567(pUZ8002)中,并利用接合转移转入吸水链霉菌Y-807中,经PCR检测、检定菌抑菌
