OBJECTIVE To explore whether and how PFC participates in the regulation of morphines effect on VTA DA neurons.METHODS & RESULTS Morphine 2 mg· kg-1 markedly excited DA cell both in firing rate and in burst.The excitation could be reversed and prevented by naloxone 0.2 mg· kg-1, suggesting its dependence on mu opiate receptor acti vation.Lesion of PFC by tetradotoxin (TTX) produced no sig nificant influence on basal DA cell activity, but abolished the morphines effect on DA cells, which indicates that the PFC is critical and might play a gating role in morphine-mediated exci tation on DA neurons.
