Indole is a privileged compound in pharmaceutical fragrance,agrochemicals,pigment and material scienceand over the past few years,[1] indole-based derivatives have been reported as highly potent agents,for example as neuropathic pain agent,anticancer agent,anti-allergic inflammatory agent,anti-HIV-1 agent,[2] and so on.There has been reported that 5-NO2 trifluoromethylated indole has excellent anti-HIV-1 activity on HIV-1 strains,[3] and many efforts have been devoted to developing efficient and selective synthetic methods to access trifluoromethylated indoles.[4] Herein we developed an efficient methodology for the synthesis of trifluoromethylated indoles using an asymmetric Friedel-Crafts alkylation of indoles with alkyl trifluoropyruvates catalyzed by our dinuclear Zinc-Catalyst and good enantioselectivities and yields were obtained.
