The purpose of the study is investigating the role of T-type Ca2+ channel in injured DRG neurons' oscillations and resonance behaviors.Whole cell patch-clamp recordings were performed on rat medium lumbar injured dorsal root ganglion (DRG) neurons exhibiting sustaining membrane potential oscillations at depolarized state.Experimental data show that firstly DRG neurons with oscillations all display resonance behaviors at depolarized state; secondly both the amplitude and the frequency of neuron's oscillations were changed in low-dose Ca2+ bath solution and neuron still exhibited oscillations in Na+-free solution at depolarization state; suggesting that Ca2+ currents are important in DRG neuron's oscillations; thirdly Ni or Amiloride abolished neuron's resonance behaviors,and Ni or Amiloride also eliminated neuron's oscillations; suggesting that T-type Ca2+ currents are essential to resonance and oscillations; Since the sustained oscillations especially near rest state are considered as the signal of super excitability correlated to the chronic neuropathic pain while T-type Ca2+ channels are essential to oscillations,in order to relieve pain the drug decreasing T-type Ca2+ current may be a good choice.
