Objective:1 Observing changes of the severity of fatty liver and abnormality of intestinal mucosal barrier in the model of nonalcoholic fatty liver disease (NAFLD) mice dynamically,to elucidate the mechanisms of the gut-liver axis in pathogenesis of NAFLD.2 To explore the effects of resveratrol (Res) on the gut-liver axis in pathogenesis of NAFLD,the levels of endotoxin,flora species of intestinal bacteria and intestinal mucosal barrier were evaluated after gavage of Res in NAFLD mice.Methods:1 48 male C57BL/6 mice,6-8 weeks,were fed by AIN93 control diet for 7 days,then were divided into normal control group (NG,n=24) and model group (MG,n=24) randomly.Two groups were given control diet plus normal drinking water,high-fat diet plus high glucose drinking water (42 g/L) respectively.Each group is divided into four time points of 4,10,18 and 24 weeks,each time point were assigned 6 mice randomly,which were recorded as NG4,NG10,NG18,NG24 and MG4,MG10,MG18,MG24 respectively;The other 18 male C57BL/6 mice,6-8 weeks,were fed AIN93 control diet for 7 days,then were assigned into normal control group (NG,n=6) and model group (MG,n=12) by 1∶2 at random.All mice were given control diet plus normal drinking water and high-fat diet plus high glucose drinking water (42 g/L) respectively.The time of model duration is 24 weeks.At the end of 24 weeks,the Control group of 6 mice were given the carboxymethylcellulosesodium (CMC) (45mg/kg/day) by gavage for 6 weeks,and 12 mice from model group were given Res or CMC (45mg/kg/day) by gavage for 6 weeks respectively,then were sacrificed at the end of 6 weeks.
