Molecular Identification of Male Sterile Cytoplasmic Types in Cabbage

  In order to identify the cytoplasmic male sterile(CMS)types in cabbage and gain the sterility-related specific sequence,the sequences of orf138 and orf224 were got by searching the nucleotide and protein databases of NCBI.Based on their conserved sequences,two pair of primers were designed and the DNA amplification of 5 CMS lines of cabbage,2 CMS lines of kohlrabi,a Brussels sprouts CMS line and their maintainer lines were carried out by PCR.The results indicated that a 749bp specific fragment was produced in all of CMS lines by amplification with the orf138 primers.The sequencing and comparison showed that the specific fragment from the 8 CMS lines and the orf138 Ogura CMS in radish reached 100%in homology,meaning they all belong to CMS type of radish.We used orf138 primers to identify the type of introduced cabbages germplasm.The eighteen materials were identified to belong to CMS type of radish.
