Using Tol2-Gal4 mediated enhancer trap to realize targeted gene expression in zebrafish

  Targeted gene expression is a powerful approach in studies of the nervous system.We have used To12 mediated enhancer trap to find tissue specific enhancers in zebrafish genome and to produce transgenic driver lines expressing Gal4 protein in specific parts of CNS.Gal4 protein can recognize upstream activator sequence (UAS) and activate the expression of UAS-driven reporter genes.Now 35 driver lines that express Gal4 in specific parts of CNS have been screened out.We have also mapped the loci of Tol2-Gal4 insertions in some of our driver lines.Some of the insertion loci are located near developmentally important genes such as zicl.These driver lines will be powerful tools for neuronal development research and behavior studies.
