Studies have found that buckwheat rutin could inhibit proliferation of cardiac fibroblasts induced by angiotensin II, but the mechanism remains uncertain.In this study, proliferation of fibroblasts model was made by adding 10 molL angiotensin Ⅱ (AngⅡ) to the medium of cultured cardiac fibroblasts of neonatal rats.Five groups were designed: control group, model group, Rutin I group, Rutin Ⅱ group and Rutin Ⅲ group.Calcineurin (CaN) activity was measured by colorimetric method, and the expression of CaN protein was observed with immunocytochemistry.Results showed Angll could remarkably increase the CaN activities and protein expression in cultured neonatal rat cardiac fibroblasts.Buckwheat rutin at dosages of 0.8 mg/L, 4.0 mg/L, 8.0 mg/L could obviously inhibit the increase of CaN activity and protein expression caused by Ang Ⅱ (P < 0.01) and showed dose-dependent relationship.The results indicated that buckwheat rutin possessed inhibitory effect on Angll - induced proliferation of fibroblasts by decreasing expression of CaN.
