TRAIL is part of the host immune system that suppresses cancer and its metastases.We have focused on unraveling mechanisms of sensitivity and resistance to TRAIL and TRAIL receptor-targeted therapeutics as we have been developing novel therapies for clinical testing.A number of mechanisms have emerged within cancer cells that appear to contribute to TRAIL resistance, and some insights have emerged regarding the potential to use TRAIL receptor targeting as therapy for cancer stem cells.The preclinical rationale for combinations of TRAIL and targeted agents will be provided in addition to new insights into resistance to TRAIL within the hypoxic tumor microenvironment.The results of high throughput screening for TRAIL sensitizers and preclinical studies implementing strategies to sensitize hypoxic tumors will be presented.The goal of this work is to unravel pathways of TRAIL signaling and resistance and to reverse the resistance in the clinic.
