NB-ARC domain proteins are one of the most diverse resistant proteins (R) in plant,which play a crucial role in effector-triggered immunity (ETI).Previous reports have indicated that some NB-ARC domain proteins were involved in responding processes of R.glutinosa consecutive monoculture practice.However, NB-ARC family proteins of R.glutinosa are little known to date.In this study, 45 NB-ARC proteins were identified from R.glutinosa protein library using hide markov model (HMM) and bioinformatics methods.Homology analysis showed that these proteins have highly homologous with Tubiflorae plants including Pedaliaceae, Scrophularuaceae, Solanaceae, Asteraceae, etc.The most of them have a typical characteristic with combining coiled-coiled domain type.A highly conserved ATP bind domain with 288 aa length containing 12 α-helix and 4 β-strand and barrel structure core of subunit enclosed by 4 parallel β-strands were observed to present in most of NB-ARC proteins by protein structure prediction.Additionally, we also found that NB-ARC domain proteins in R.glutinosa had strong homology to the R1A and RGA proteins, which participated to reorganization of Avrl effectors and salicylic acid-mediated signal transduction.Most of the closely related members in the phylogenetic tree exhibited common motifs as expected.The expression patterns analysis of 36 NB-ARC family proteins indicated that the effective immune resistances were triggered approximately at 30 days after planting in R.glutinosa.Then it might be more sensitive period to induce occurrence of consecutive monoculture problems at the next 30 days.
