Glycine is a principal inhibitory neurotransmitter in the vertebrate retina.Using immunocytochemical and whole cell recording techniques,we investigated the expression of the glycine transporter 1 (GlyT1) on bullfrog retinal photoreceptors.Double labeling of recoverin,a specific marker for photoreceptors,and GlyT1 showed that labeling for GlyT1 was seen in the outer nuclear layer (ONL) and outer plexiform layer (OPL).No labeling for GlyT2 was found in the bullfrog retina.Furthermore,both the terminals and inner segments of isolated cone photoreceptors were labeled by GlyT1.In retinal slice preparation,we further found that local application of glycine induced an inward current from cones,which was only in part suppressed by strychnine (10 μM),and the remaining strychnine-resistant component was blocked by NFPS,a specific GlyT1 antagonist.Puffing sarcosine,an agonist of GlyT1,induced a similar inward current.These results suggest that GlyT1 is functionally expressed on bullfrog cones,which may be involved in information processing in the OPL.
