The impacts of high fat diet (HFD) induced maternal obesity that prone the offspring to metabolic diseases are well documented.However, the transgenerational effects of continuous HFD feeding of offspring over one generation are still unknown.Previously, we demonstrated transgenerational exacerbation of fatty liver disease in male offspring if their grandmother and mother were fed with HFD, which suggested a "feed-forward cycle" existed.Here, we demonstrated that this "feed-forward cycle" also happened on the adipose tissue of female mice.C57BL/6 mice were fed with HFD or normal chow for three generations, named F0, F 1 and F2.Gradually increased body weight and fat weight but not the size of adipocytes were observed in female offspring under continuous HFD feeding.Microarray and qRT-PCR revealed that the transgenerational changed genes were mainly involved in the inflammatory responses.Bisulfite sequencing revealed that the promoters of TLR1, TLR2 and Lat exhibited hypomethylation in the F2 generation.Our data demonstrated the female offspring exhibited a transgenerational increase in inflammatory status in the adipose tissue due to DNA hypomethylation under the continuous HFD feeding.
