Transcription activator-like effector nucleases (TALENs) are among the most efficient genome-editing tools.β-Lactoglobulin (BLG) is a major whey protein in cow and goat milk and has been demonstrated to be an important milk allergen.Unlike the prime pharmaceutical interest to generate animal bioreactors in livestock, our intent is to produce transgenic goats with human α-lactalbumin (hLA)-enriched whey proteins and reduced BLG content in their milk.In the present study, we inserted the hLA gene into the goat BLG exonl locus by TALEN-mediated homologous recombination (HR) and we expected that endogenous BLG 5regulatory element could promote hLA expression.
