Objective: The aim of this study was to vividly display the revascularization mechanism of both pedicle flaps and free flaps by reciprocal pedicle flaps and reciprocal free flaps model, respectively.Bone marrow mesenchymal stem cells (BMSCs) were transduced by stromal cell-derived factor (SDF), in order to assess its ability to promote neovascularization of both flaps on the basis of revascularization mechanisms or not?Methods: The reciprocal pedicle flaps model was transferred between green fluorescent protein (GFP) transgenic C57BL/6 mice and wild-type C57BL/6 mice.The reciprocal free flaps model was established between sex mixmatched wild-type Lewis rats.Then blood vessels source, growth direction and anastomosis position were detected by fluorescence microscope, immunohistochemistry, fluorescence angiography and FISH at postoperative 3, 7, 14 and 21 days, respectively.The Ad-SDF-1α transduced and untransduced BMSCs and Ad-SDF-1α were transplanted into pedicle flaps and free flaps, respectively.The blood vessel growth, vascular density and flap survived areas were assessed by HE staining, immunohistochemistry and other methods at postoperative 7 and/or 10 days.
