Aim: To investigate whether lentivirus (LV)-mediated platelet-specific gene therapy will correct murine hemophilia B phenotype and induce immune tolerance in factor Ⅸ-deficient (FⅨnull) mice.Methods: The 2bF9 lentiviral vector (pWPT-2bF9) harboring human FⅨ expression cassette under the control of the platelet-specific glycoprotein Ⅱb gene (? Ⅱb) promoter was used in this study.Six to 8 week old FⅨnull mice were conditioned for cellular transplantation with 660 or 1100 cGy total body irradiation.
