Objective To investigate the inhibiting effect of salidroside on human gastric carcinoma cells SGC-7901 and its mechanism.Methods Human gastric carcinoma cells SGC-7901 were divided into four groups: 0.5 mg/mL salidroside group, 1.0 mg/mL salidroside group, 2.0 mg/mL salidroside group and control group.The proliferation of cells were assayed by MTT assay and argyrophilic staining (AgNORs).Flow cytometry was used to assess the cell cycle progression and apoptosis.The migration activity of gastric carcinoma cells SGC-7901 was detected by cell scratch assay.Results MTT assay results showed that the the salidroside drug concentration group can inhibit the growth and proliferation of gastric carcinoma cells SGC-7901, and the inhibition rate was significantly higher than control group (P<0.05).Argyrophilic staining experiments showed that: AgNORs values in salidroside group (0.5 mg/mL, 1.0 mg/mL and 2.0 mg/mL) were (2.91±0.74) / nuclear, (2.35±0.48)/nuclear and (1.68±0.67)/nuclear respectively, which significantly lower than those in the control group (3.80±0.l) / nuclear.Proliferation of gastric carcinoma cells SGC-790 was inhibited (P<0.05).In salidroside groups, the proportion of gastric carcinoma cells SGC-7901 in S phase was higher and the proportion of cells in G2/M phase was lower than that in control group (P<0.05).The apoptotic rates of gastric carcinoma cells SGC-7901 in 0.5 mg/ml salidroside group, 1.0 mg/ ml salidroside group and 2.0 mg/ml salidroside group were (25.95±0.10)%, (29.64±0.11)% and (31.72±1.18)%respectively, which were higher than that in control cells (8.20±0.10)% (P<0.05).The relative migrations of gastric carcinoma cells SGC-7901 which were incubated with 0.5 mg/mL, 1.0 mg/mL and 2.0 mg/mL salidroside after 48 h were(72.60±0.50)%, (65.26±1.46)% and (59.83±3.18)% respectively, which were less than that of control cells (81.38±0.46)% (P<0.05).Conclusion Salidroside can effectively inhibit growth and proliferation of gastric carcinoma cells SGC-7901, and induce apoptosis and reduce the migration.The inhibiting effect on growth and proliferation of human gastric carcinoma cells SGC-7901 enhanced with the increased concentration of salidroside.
