A novel macromolecular magnetic resonance imaging ( MRI ) contrast agent constituted of arabinogalactan derivative conjugated with gadolinium tetraazacyclododecanetetraacetic acid ( Gd-DOTA) was synthesized and characterized.The T1-relaxivities of 7.6 mM-1 s-1 for Gd-DOTA-AGCMC6 was much higher than that of a commercial MRI contrast agent( Gd-DOTA) in D20 at 25 ℃ and 9.4 T.It was 7.8 mM-1 s-1 for Gd-DOTA-AGCMC6 in 0.725 mM BSA solution,which was l.03 times that of Gd-DOTA-AGCMC6 in D20.It was deduced that about l.8% of Gd-DOTA-AGCMC6exist as Gd-DOTA-AGCMC6 BSA under the condition of [ BSA] =0.725 mM.MR imaging of Wistar rats showed remarkable enhancement in rat liver and kidney after Gd-DOTA-AGCMC6 injected,which persisted longer than Gd-DOTA.The mean percentage enhancement( Gd-DOTA-AGCMC6 at gadolinium dose of 0.086 + 0.002 mmol/kg) was as following,liver parenchyma at 10-40 min: 46.17 + 8.15% ; kidney at 10-30 min: 140.77 + 18.56%.The present investigation indicates that arabinogalactan-bound complexes are potential liver-specific contrast agents for MRI.
