Fluoroquinolones(FQs)are a widely used class of synthetic antibiotics with broad activity spectrum of mycoplasma and bacteria(both Gram positive and negat ive).Due to the extensive use of FQs in veterinary science and pharmaceutical industry,they are expected to enter the environmental water via animal excretion and hospital waste,giving rise to a constant need for a sensitive and reliable method to monitor FQs in aquatic environment.Thus,an off-line procedure of immunoaffinity extraction(IAE)followed by capillary electrophoresis-mass spectroscopy(CE-MS)has been developed for simultaneous determinat ion of eight typical FQs in environment water.A broad-specific polyclonal antibody was home-prepared and immobilized on a Sepharose 4B stationary phase for sample cleanup,FQs extraction and pre-concentration.CE-MS was subsequently applied for FQs separation and quantification.The immunoaffinity column was evaluated in the parameters of selectivity,binding capacity,elution protocol,and recovery.The separation and detection by CE-MS were also investigated.Under the optimized conditions,the developed method was finally applied to analyze FQs in environmental water sample from a local lake,providing satisfactory recoveries from 72%to 112%and LODs ranging from 1.2 nM to 5.0 nM.
