Objective Astrocyte becomes active in response to CNS injury.However, the mechanism of astrocytic activation is largely unknown.In this study, we explored the roles of TRPC on astrocyte activation induced by lipopolysaccharide (LPS) in vitro.Methods we treated cultured rat neonatal cortical astrocyte with LPS to establish a model of astrocytic activation.Then, the expression of TRPC mRNA were detected by real-time PCR and the concentration of intracellular calcium([Ca2+]i) by laser scanning confocal microscopy.Pharmacological characterizations of TRPC on astrocytic activition were observed with two kinds of TRPC blockers.Results LPS, a well-known inflammatory stimulus, induced astrocytic activation and increases in TRPC3 and TRPC6 mRNA levels and [Ca2+]i.Furthermore, TRPC antagonists, 2-APB and SKF96365, inhibited LPS-induced cell proliferation, the increase in glial fibrillary acidic protein expression and the release of pro-inflammatory cytokines, IL-6 and IL-1β.Meanwhile, we found that 2-APB and SKF96365 inhibited an increase of [Ca2+]i.Conclusion TRPC, especially TRPC3 and TRPC6 are involved in astrocytic activation.
