Divalent metal transporter 1 (DMT 1) is responsible for ferrous-ion transport. The over expression of DMT 1 in the brain has been suggested to be involved in iron accumulation in some neurodegenerative disorders. The aim of the present study is to construct two forms of DMT1 (with IRE and without IRE) over expression model to investigate the relationship between increased DMT1 expression and iron accumulation in MES23.5 cells, hence illustrating over expressed DMT1 resulted cell dysfunction.RT-PCR and western blot confirmed the increased DMT1 expression in AdDMT1+IRE and AdDMT1-IRE infected MES23.5 cells.
