Protective Effects and Action Mechanisms of Oxysophoridine on Anoxia Injury in Primary Cultured Rat

　　Objective: To investigate the protective effects and action mechanism of Oxysophoridine (OSR) on primary cultured hippocampal neurons subjected to anoxia injury in rat.Methods: Anoxia injury ofhippocampal neurons that cultured in vitro were produced by physical oxygen deficiency and using culture fluid without Glu.The survival rate of neurons, The leaking rate of lactate dehydrogenase (LDH), the contents of malondialdehyde (MDA) and nitric oxide (NO), the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) and nitric oxide synthase (NOS) were measured.The intracellular free calcium concentration ([Ca2+]i) in hippocampal neurons were measured by using Ca2+-sensitive fluorescent probe Fluo-3/AM with dual wavelength fluorescence spectrophotometer.Results: The model of anoxia injury can due to cell death markedly and arise the leaking rate of LDH.The model can increase the contents ofNO,MDA, [Ca2+]i and the activity of NOS, also inhibit the increasing of SOD, GSH-PX activities.OSR (0.625,5,10 ug/L)can reduced the hippocampal neurons subjected to anoxia injury.Conclusion: OSR has significant protective effects on hippocampal neurons subjected to anoxia injury.The effective mechanism of OSR might relate to anti-oxidation and calcium overload.
