The binding of amino containing compounds such as neomycin,polyamidoamine dendrimer (PAMAM),polyacrylic amine (PAH) and polyacryl acid (PAA) to TAR RNA immobilized on a biosensor surface has been studied using microgravimetric quartz crystal microbalance (QCM).TAR RNA was connected with biotin and fixed to the avidin modified QCM surface via the avidin-biotin interaction,so that resulted in a TAR RNA coated QCM surface.Experimental results showed that many amino containing compounds could form complexes with TAR RNA,especially,PAMAM dendrimer that could disrupt the interaction of Tat-peptide with TAR RNA,which is essential for HIV-1 virus replication.An equation similar to Langmuir Isotherm is suggested for quantitatively measuring the ‘surface binding constant’ between TAR RNA on the substrate and target molecule in solution.
