Glycosylation is the most common and important post-translational modification of proteins.Glycopeptides enrichment is important for glycoproteomics because of low concentration of glycopeptides and suppression effect of abundant non-glycosylated peptides in mass spectrometry. In recent years, hydrophilic interaction chromatography (HILIC) in solid phase extraction (SPE) mode has been increasingly employed to enrich glycopeptides.TiO2 under HILIC mode exhibited higher enrichment selectivity for glycopeptides than Sepharose. However, this method was not sufficient uniform to enrich glycopeptides from unknown samples because of different loading, washing and elution conditions for different glycopeptides.
