Sodium houttuyfonate (SH) is a compound,it is consists of sodium bisulfite and houttuynin.Houttuynin is one of the key components in the essential oil of houttuynia cordata thunb, which has been widely used to cure phlogosis as an anti-inflammatory agent,although the mechanisms are not fully understood yet.The aim of this study was to investigate the anti-inflammatory potential of SH and its mechanisms of action in vitro.Monolayer primary bovine endometrial epithelial cells (bEECs) were stimulated with lipopolysaccharide (LPS) in the presence or absence of SH.The anti-inflammatory effects of SH were measured by production of inflammatory mediators by quantitative real-time polymerase chain reaction (qRT—PCR) and ELISA, as well as expression of Toll-like receptor 4 (TLR4) ,inhibitor of kappa B (IκB)α and phosphor—IκBα kinase (p—IKK) by western blot.Treatment of bEECs with SH significantly inhibited all of the inflammatory parameters measured,including LPS—induced Interleukin (IL)—6 and IL.—8 production, TLR4 expression, IκBα degradation and IKK phosphoryiation.In a LPS—induced inflammation of cell model,SH inhibited the production of f.umor necrosis factor (TNF)—α, IL.—1 β, IL—6 and IL—8 on mRNA level.SH possesses potent anti-inflammatory activity in vitro ,which occurs at least partly through inhibition of pro-inflammatory signaling and mediator release.These results strongly support the therapeutic potential of SH might inhibit the expression of inflammatory cytokines through inactivation of nuclear factor kappa B (NF—κB) via reducing IKK phosphorylation and IκBα degradation.
