粗线期piRNA指导小鼠精子形成后期mRNA的大规模清除

  在哺乳动物的精子发生过程先后出现两次piRNA的表达高峰,分别被称为前粗线期piRNA与粗线期piRNA.前粗线期piRNA主要负责沉默逆转座子等可移动性元件,而主要来源于基因间区域的粗线期piRNA的功能现在还知之甚少.我们发现,在小鼠延长形精子细胞中,粗线期piRNA与其结合蛋白MIWI、脱腺苷酸酶CAF1组成pi-RISC复合物,通过碱基不完全配对方式识别靶mRNA 3非翻译区(3 UTR)的序列元件、诱导靶mRNA脱腺苷酸及降解;依赖于piRNA百万级数量的不同序列,pi-RISC介导了精子细胞发育后期数千不同mRNA的降解.这一研究工作不仅提供了精子形成后期mRNA大规模降解的分子机制,同时揭示了粗线期piRNA在精子发育中的重要功能,并证明piRNA除了沉默转座子外,还参与调控编码基因.
